Indication:

Vorasidenib is used to treat patients 12 years and older with a type of slow-growing brain tumor called Grade 2 astrocytoma or oligodendroglioma that carries an IDH1 or IDH2 gene mutation and has not progressed after surgery. Dosage:                               

Vorasidenib (brand name Voranigo) is an oral targeted therapy for patients aged 12 and older with Grade 2 astrocytoma or oligodendroglioma that have specific IDH1 or IDH2 mutations, after initial surgery. The standard adult dose is 40 mg once daily, taken by mouth with or without food, and continued until the disease progresses or toxicity becomes unacceptable. For adolescents (≥12 years), dosing is based on body weight: those weighing ≥ 40 kg receive 40 mg daily, while those < 40 kg get 20 mg daily. Tablets must be swallowed whole—not crushed, chewed, or split—with missed doses taken within 6 hours or skipped if later.

Note:

Liver function tests (AST/ALT) are required before starting treatment, every 2 weeks for the first 2 months, then monthly for up to 2 years .

If liver enzymes rise significantly, dose reductions are recommended: first to 20 mg, then to 10 mg daily; if 10 mg is not tolerated or toxicity persists, therapy should be stopped .

Safe use requires monitoring kidney function and advising on contraception, since it may harm unborn babie.
