Indication:

Neratinib is used to treat early-stage HER2-positive breast cancer in adults, especially after completing treatment with trastuzumab (another HER2-targeted therapy). It helps lower the risk of the cancer coming back by blocking HER2 receptors, which are proteins that help cancer cells grow. Neratinib is often used as an extended adjuvant therapy, meaning it continues treatment after initial therapy to give longer protection against recurrence.

Dosage:

Neratinib is usually taken as 240 mg once daily (which means six 40 mg tablets at the same time) with food, for up to 1 year, depending on the doctor’s plan. It is mainly prescribed for early-stage HER2-positive breast cancer after completing other treatments like trastuzumab. To reduce side effects like diarrhea, patients often start with an anti-diarrheal medicine such as loperamide, especially during the first 1–2 months of treatment.

Note:

Take the tablets with food, at the same time each day.

Do not crush or split the tablets.

Diarrhea is a common side effect, so staying hydrated and following your doctor’s instructions on managing it is important.

Regular monitoring is needed to check for liver function and side effects.

Side Effect:

The most common side effects of Neratinib are:

Nausea, vomiting, headache, tiredness, constipation, and diarrhea. Tell your doctor or pharmacist right away if any of these side effects persist or worsen.

Remember that your doctor has prescribed this medication because your doctor has judged that the benefit to you is greater than the risk of side effects. Many people using this medication do not have serious side effects.

Tell your doctor right away if you develop diarrhea that is severe or doesn't stop, or signs of dehydration (such as unusual decreased urination, unusual dry mouth/thirst, fast heartbeat, dizziness/lightheadedness). Your doctor may prescribe additional anti-diarrhea medications or temporarily stop/adjust your dose of neratinib.

This is not a complete list of possible side effects. Contact your doctor or pharmacist if you have any other side effects not listed above.
